Sixty-nine per cent of students who sat the assessment passed, compared with 65% who passed in September 2018.
A total of 1,057 candidates sat the exam on September 26, of which 731 passed, the regulator said.
GPhC chief executive Duncan Rudkin congratulated those who passed, adding that he looks forward to them joinining the register.
"Assessments are carefully set so that only the candidates who demonstrate the required knowledge and understanding can pass and be eligible to join the register," he said.
The pass rate for the June pre-reg exam dropped to 72%, the lowest June result since the GPhC took over responsibility for the assessment in 2011.
